RESUMO

N-acyl homoserine lactones (AHLs) function as signaling molecules influencing microbial community dynamics. This study investigates the impact of exogenously applied AHLs on methane production during waste-activated sludge (WAS) anaerobic digestion (AD). Nine AHL types, ranging from 10-4 to 10 µg/g VSS, were applied, comparing microbial community composition under optimal AHL concentrations. Firmicutes, Bacteroidetes, Chloroflexi, and Proteobacteria were identified in anaerobic digesters with C4-HSL, C6-HSL, and C8-HSL. Compared to the control, Halobacterota increased by 19.25%, 20.87%, and 9.33% with C7-HSL, C10-HSL, and C12-HSL. Exogenous C7-HSL enhanced the relative abundance of Methanosarcina, Romboutsia, Sedimentibacter, Proteiniclasticum, Christensenellaceae_R-7_group. C10-HSL increased Methanosarcina abundance. C4-HSL, C6-HSL, C8-HSL, C10-HSL, and C12-HSL showed potential to increase unclassified_Firmicutes. Functional Annotation of Prokaryotic Taxa (FAPROTAX) predicted AHLs' impact on related functional genes, providing insights into their role in AD methanogenesis regulation. This study aimed to enhance the understanding of the influence of different types of exogenous AHLs on AD and provide technical support for regulating the methanogenesis efficiency of AD by exogenous AHLs.

This study aimed to improve anaerobic digestion (AD) efficiency through the addition of zero-valent iron (ZVI) and biogas slurry. This paper demonstrated that methane production was most effectively promoted at a biogas slurry reflux ratio of 60%. The introduction of ZVI into anaerobic systems does not enhance its bioavailability. However, both biogas slurry reflux and the combination of ZVI with biogas slurry reflux increase the relative abundance of microorganisms involved in the direct interspecific electron transfer (DIET) process. Among them, the dominant microorganisms Methanosaeta, Methanobacterium, Methanobrevibacter, and Methanolinea accounted for over 60% of the total methanogenic archaea. The Tax4Fun function prediction results indicate that biogas slurry reflux and the combination of ZVI with biogas slurry reflux can increase the content of key enzymes in the acetotrophic and hydrotrophic methanogenesis pathways, thereby strengthening these pathways. The corrosion of ZVI promotes hydrogen production, and the biogas slurry reflux provided additional alkaline and anaerobic microorganisms for the anaerobic system. Their synergistic effect promoted the growth of hydrotrophic methanogens and improved the activities of various enzymes in the hydrolysis and acidification phases, enhanced the system's buffer capacity, and prevented secondary environmental pollution. PRACTITIONER POINTS: Optimal methane production was achieved at a biogas slurry reflux ratio of 60%. Biogas slurry reflux in anaerobic digestion substantially reduced discharge. ZVI addition in combination with biogas slurry reflux facilitates the DIET process.

The effective inhibition of nitrite-oxidizing bacteria (NOB) is widely acknowledged to be a critical issue for mainstream short-cut biological nitrogen removal. This study demonstrated a stable mainstream nitritation by implementing light irradiation. A sequencing batch reactor with ultraviolet-A (UVA) irradiation was operated for 250 days, and a high nitrite accumulation ratio was achieved and stabilized at about 90 %. UVA irradiation also positively impacts denitrification activity, with total nitrogen removal up to 63 %. Microbial community analysis confirmed that the UVA effectively and stably decreased the abundance of Nitrospira (the only detected NOB) from 6.0 % to 0.1 %, while it showed no effect on Nitrosomonas. The enriched genus Rhodocyclaceae was the major contributor to the increase in denitrification activity in the light-induced nitritation system. The proposed UVA irradiation strategy has the potential to be integrated with an anoxic/aerobic (A/O) or integrated fixed-film activated sludge (IFAS) process for achieving mainstream short-cut biological nitrogen removal.

To assess the problem of sewage treatment under the condition of low carbon sources, we carried out a study of activated sludge and a biofilm symbiosis system (IFAS). The occurrence characteristics and interaction law of microorganisms in two phases of sludge membrane under low carbon source conditions were discussed, and their niche and influence on treatment efficiency were clarified. Through a pilot-scale experiment in actual water plants, the biofilm characteristics, sludge membrane activity, and succession law of flora were analyzed, and the microbial structure and interaction in sludge membrane in two phases under the control of different activated sludge ages were compared. The results showed that the sludge concentration in the reactor increased with the increase in SRT under variable SRT. Because the microbial concentration in SRT-H was much higher than that in SRT-L, the competition between mud films in SRT-H was more intense than that in SRT-L, and the pollutant removal efficiency in SRT-H was lower than that in SRT-L. Under the condition of low-carbon feed water, the sludge activity in the IFAS process decreased with the increase in SRT. Under the condition of low SRT(5 d), the nitrification, denitrification, phosphorus accumulation, and phosphorus absorption rate of activated sludge increased by 122%, 88%, 34%, and 44%, respectively, compared with that of high SRT (25 d). However, SRT had little effect on biofilm activity, and there was little difference in nitrification activity and denitrification activity between the two SRTs. Microbial sequencing analysis showed that the functional bacteria of the IFAS process were enriched and transferred with the change in SRT between the two phases of mud membrane. In SRT-L, the functional bacteria that were enriched and transferred between the two phases of mud film owing to the "seeding" effect were mainly unclassified_g__Enterobacteriaceae, whereas in SRT-H, Acinetobacter was mainly used. At the same time, by analyzing the distribution of dominant functional bacteria, it was found that there was some competition between denitrifying bacteria and phosphorus-accumulating bacteria in activated sludge. Under the condition of a lack of organic substrate in the influent, the relative abundance of denitrifying bacteria was obviously higher than that of phosphorus-accumulating bacteria, which indicated that denitrifying bacteria could better adapt to low-carbon source conditions. Thus, they could occupy a dominant competition position, which was mainly reflected in the increase in the relative abundance of aerobic denitrifying bacteria. In addition, the SRT change in the mud phase reacted in the membrane phase, making the residence time of biofilm change correspondingly, thus changing the flora structure, screening out different dominant bacteria genera, and further increasing the difference.

In the present study, glutaraldehyde was used as a hydrophobic modifier to crosslink polyvinyl alcohol (PVA), and copper ion was immobilized by sodium alginate (SA). Polyvinyl alcohol-copper alginate (PVA-CA) gel beads were prepared by a one-step process, and were used to adsorb and remove tetracycline (TC) from an aqueous solution. The beads were characterized by scanning electron microscopy (SEM), thermogravimetric analysis (TGA), Brunauer-Emmett-Teller (BET) measurement, X-ray diffraction (XRD), energy dispersive spectroscopy (EDS), and Fourier transform infrared spectroscopy (FTIR). The adsorption experiment showed that the optimal pH value of the beads was 5, and that their adsorption met pseudo-second-order kinetic and Langmuir isothermal models. The adsorption thermodynamics experiment showed that the adsorption process was spontaneous and endothermic. Under optimal adsorption conditions, the maximum adsorption capacity for TC of the beads was 231.431 mg/g, which was much higher than that of a single copper alginate matrix. After 5 adsorption-desorption cycles, the adsorption capacity remained high. FTIR and X-ray photoelectron spectroscopy (XPS) revealed that the cation bonding bridge reaction was the main driving force behind the adsorption mechanism. Compared with other reported adsorption materials, the PVA-CA gel beads have high adsorption capacity, a simple preparation process, and excellent recovery performance.

This first-attempt study illustrated the microbial cooperative interactions related to bioelectricity generation from the mixture of sludge fermentation liquid (SFL) and fruit waste extracts (FWEs) via microbial fuel cells (MFCs). The optimal output voltages of 0.65 V for SFL-MFCs, 0.51 V for FWEs-MFCs and 0.75 V for mixture-MFCs associated with bioelectricity conversion efficiencies of 1.061, 0.718 and 1.391 kWh/kg COD were reached, respectively. FWEs addition for substrates C/N ratio optimization contributed considerably to increase SFL-fed MFCs performance via triggering a higher microbial diversity, larger relatively abundance of functional genes and microbial synergistic interactions with genera enrichment of Clostridium, Alicycliphilus, Thermomonas, Geobacter, Paludibaculum, Pseudomonas, Taibaiella and Comamonas. Furthermore, a conceptual illustration of co-locating scenario of wastewater treatment plant(s), waste sludge in situ acidogenic fermentation, fruit waste collection/crushing station and MFC plant was proposed for the first time, which provided new thinking for future waste sludge treatment toward maximizing solid reduction and power recovery.

Tetracycline (TC) is a commonly used human and veterinary antibiotic that is mostly discharged into wastewater in the form of the parent compounds. At present, wastewater treatment plants (WWTPs) use activated sludge processes that are not specifically designed to remove such pollutants. Considering the biological toxicity of TC in aquatic environment, the migration and fate of TC in the process of wastewater treatment deserve attention. This paper reviews the influence of TC on the functional bacteria in the sludge matrix and the development of tetracycline-resistant genes, and also discusses their adsorption removal rates, their adsorption kinetics and adsorption isotherm models, and infers their adsorption mechanism. In addition, the biodegradation of TC in the process of biological treatment is reviewed. Co-metabolism and the role of dominant bacteria in the degradation process are described, along with the formation of degradation byproducts and their toxicity. Furthermore, the current popular integrated coupling-system for TC degradation is also introduced. This paper systematically introduces the interaction between TC and activated sludge in WWTPs. The review concludes by providing directions to address research and knowledge gaps in TC removal from wastewater.

Nitritation is currently known as a bottleneck for mainstream nitrite shunt or partial nitritation/anammox (PN/A). Here we propose a new approach to selectively eliminate nitrite oxidizing bacteria (NOB) for mainstream nitritation by low-dose ultraviolet-A (UVA) irradiation. The results showed that mainstream nitritation was rapidly achieved within 10 days with UVA irradiation at the dose of 0.87 µE L-1 s-1, and nitrite accumulation ratio (NO2--N/(NO2--N + NO3--N) ×100%) stabilized over 80%. Microbial community analysis revealed that two typical NOB populations (Nitrospira and Ca. Nitrotoga) detected in the control reactor were suppressed efficiently in UVA irradiation reactor, whereas the Nitrosomonas genus of ammonium oxidizing bacteria (AOB) remained at similar level. Intracellular reactive oxygen species (ROS) analysis indicated that NOB-dominant sludge tends to generate more intracellular ROS compared with AOB-dominant sludge in the presence of UVA, leading to the inactivation and elimination of NOB. Additionally, amounts of microalgae found in UVA irradiation reactor could help to suppress NOB by generating ROS during photosynthesis. Briefly, the UVA irradiation approach proposed in this study was shown to be promising in NOB suppression for reliable mainstream nitritation.

Effect of temperature downshifts on process performance and bacterial community dynamics was investigated in a bench-scale hybrid A/O system treating real domestic wastewater. Results showed that the average COD removal in this system reached 90.5%, 89.1% and 90.3% for Run 1 (25 °C), Run 2 (15 °C) and Run 3 (10 °C), respectively, and variations in temperature barely affected the effluent COD concentration. The average removal efficiencies of NH4(+)-N were 98.4%, 97.8%, 95.7%, and that of TN were 77.1%, 61.8%, 72% at 25 °C, 15 °C and 10 °C, respectively. Although the hybrid system was subjected to low temperature, this process effectively removed NH4(+)-N and TN even at 10 °C with the average effluent concentrations of 2.4 mg/L and 14.3 mg/L, respectively. Results from high-throughput sequencing analysis revealed that when the operation temperature decreased from 25 °C to 10 °C, the richness and diversity indexes of the system decreased in the sludge samples, while underwent an increase in the biofilm samples. Furthermore, the major heterotrophic bacteria consisted of Lewinella, Lutimonas, Chitinophaga and Fluviicola at 10 °C, which could be central to effective COD removal at low temperature. Additionally, Azospira, one denitrifying-related genus increased from 0.4% to 4.45% in the biofilm samples, with a stable TN removal in response to temperature downshifts. Nitrosomonas and Nitrospira increased significantly in the biofilm samples, implying that the attached biofilm contributed to more nitrification at low temperature.

A biofilter was developed in this study, which showed an excellent performance with the simultaneous removal of AsIII from 150 to 10mg L(-1) during biological iron and manganese oxidation. The distribution and genetic diversity of the microorganisms along the depth of the biofilter have been investigated using DGGE. Results suggested that Iron oxidizing bacteria (IOB, such as Gallionella, Leptothrix), Manganese oxidizing bacteria (MnOB, such as Leptothrix, Pseudomonas, Hyphomicrobium, Arthrobacter) and AsIII-oxidizing bacteria (AsOB, such as Alcaligenes, Pseudomonas) are dominant in the biofilter. The spatial distribution of IOB, MnOB and AsOB at different depths of the biofilter determined the removal zone of FeII, MnII and AsIII, which site at the depths of 20, 60 and 60cm, respectively, and the corresponding removal efficiencies were 86%, 84% and 87%, respectively. This process shows great potential to the treatment of groundwater contaminated with iron, manganese and arsenic due to its stable performance and significant cost-savings.

In iron and manganese-containing groundwater treatment for drinking water production, biological filter is an effective process to remove such pollutants. Until now the exact microbial mechanism of iron and manganese removal, especially coupled with other pollutants, such as ammonia, has not been clearly understood. To assess this issue, the performance of a full-scale biofilter located in Harbin, China was monitored over four months. Microbial populations in the biofilter were investigated using T-RFLP and clone library technique. Results suggested that Gallionella, Leptothrix, Nitrospira, Hyphomicrobium and Pseudomonas are dominant in the biofilter and play major roles in the removal of iron, manganese and ammonia. The spatial distribution of microbial populations along the depth of the biofilter demonstrated the stratification of the removal of iron, manganese and ammonia. Additionally, the absence of ammonia-oxidizing bacteria in the biofilter implicated that ammonia-oxidizing archaea might be responsible for the oxidation of ammonia to nitrite.
